精神分裂症患者血清中发现特征蛋白

摘　　要：目的:采用蛋白组学高灵敏高通量研究工具―表面增强激光解析电离飞行时间质谱(SELDI-TOF-MS)技术检测血清蛋白指纹图,通过差异蛋白组学筛选精神分裂症特有的蛋白标记物,为研究精神分裂症的病理过程和实验诊断奠定基础。方法:用Ciphergen公司生产的PBSⅡ/C型蛋白质指纹图谱仪和ProtienChip CM10(弱阳离子交换表面)蛋白芯片采集32例精神分裂症和44例对照组血清蛋白质谱图,采用Biomaker Wizard3.1版分析软件筛选差异蛋白和Biomaker pattern software5.0.2版建立诊断模型。结果:发现精神分裂症患者与对照组血清蛋白质指纹图谱有115个差异表达的蛋白质,其中6个有明显表达差异的标志蛋白,其质荷比(M/Z)分别为2821、3 219、3317、4284、4347和4357,建立诊断模型对精神分裂症的诊断灵敏度和特异性分别为93.75%和93.18%,阳性预测值和阴性预测值分别为90.09%和95.35%。结论:精神分裂症患者血清具有高表达的特征蛋白,对精神疾病的鉴定、诊断、治疗和基因研究具有重要意义。Objective：To develop a novel and reliable method for diagnosis of schizophrenia by surface enhanced laser desorption/ionization time-of-flight mass spectrometry （SELDI-TOF-MS）.Methods ：The study ineluded 32 patients with schizophrenia and 44 controls （include 28 healthy volunteers and 16 other diseases）. For serum protein signature anal）isis, the SELDI-TOF-MS analysis was performed to screen the serum protein markers and the CM10 （weak cation exchange array） protein chips were used for analysis.A diagnostic model was developed with Biomaker pattern software5.0.2. Results ：The data analysis revealed 115 differential expression peaks. Six specific protein peaks with M/Z at 2 821,3 219,3 317,4 284,4 347,4 357,were chosen to develop a diagnostic model. When the model was blindly tested with the test set, it yielded a sensitivity of 93.75%,a specificity of 93.18%,and a negative and positive predictive value respectively of 95.35% and 90.09%. Conclusion： Differential proteins were detected from serum of Schizophrenia. Combination of SELDI-TOF-MS with CM10 protein chips may provides a new proteomics mode for laboratory diagnosis of schizophrenia. These specific biomarkers is a potential tool for early diagnosis and identification of schizophrenia.
