p38激活丝裂原活化蛋白激酶和转化生长因子-β_1在糖尿病大鼠肾小管间质表达的动态观察

摘　　要：①目的探讨p38激活丝裂原活化蛋白激酶(p38MAPK)与转化生长因子-β1(TGF-β1)在肾小管间质病变发生、发展中的作用。②方法用W istar大鼠建立链脲佐菌素(STZ)诱导的糖尿病(DM)大鼠模型,设正常对照组(A)和糖尿病组(B)。用免疫组化和RT-PCR方法观察肾小管间质p38MAPK和TGF-β1蛋白及其mRNA的表达;HE和PAS染色,光镜观察动物肾小管形态学改变;生化法测定血糖、尿素氮、血肌酐及24小时尿蛋白。③结果p38MAPK和TGF-β1在DM大鼠肾小管间质的表达较对照组显著升高(P<0.05)。④结论糖尿病状态下,p38MAPK和TGF-β1在大鼠肾小管间质活性明显增高,它们可能共同参与了糖尿病大鼠肾小管间质损害的过程。Objective To investigate the expression of p38 mitogen - actvated protein kinase （ p38 MAPK） and transforming growth factor - β1 （ TGF - β1 ） in the tubulointerstitium of diabetic rats. Methods Streptozotocin - induced diabetic wistar rats were divided into the control group A and diabetic group B. The changes of expressions of p38 MAPK and TGF - β1 proteins were analyzed with the routine immunohistochemical method. At the same time, expressions of p38 MAPK and TGF - β1 mRNA were detected by RT - PCR. The morphology of renal interstitium was examined with HE and PAS stain. Blood glucose, blood serum creatinine, blood urea nitrogen and 24h urinary protein were examined with biochemical methods. Results The levels of p38 MAPK and TGF - β1 in group B were significantly higher than those in group A （ P 〈 0.05 ）. Conclusions mRNA and protein of p38 MAPK and TGF - β1 were over - expressed in the tubulointerstitium of diabetic rats induced by STZ and contributed to the pathogenesis of the tubulointerstitial injury.
