摘 要:目的组装癌症治疗药物五氟尿嘧啶,构成功能性磁性壳聚糖纳米粒子药物载体。方法用化学共沉淀法制备了Fe3O4纳米颗粒,经优化胶体化学反应技术采用壳聚糖改性。经激光粒度仪、透射电子显微镜,红外光谱以及紫外-可见光谱进行形貌与粒度研究分析。结果其中磁性Fe3O4颗粒,壳聚糖改性磁性Fe3O4,以及载药(五氟尿嘧啶,5-Fu)壳聚糖改性磁性Fe3O4纳米颗粒的粒径分别为8 ̄12nm,12 ̄15nm和20 ̄30nm,颗粒形状呈规则球型;壳聚糖改性作用和改性后纳米磁性Fe3O4载药效果明显。壳聚糖-五氟尿嘧啶(5-Fu)和壳聚糖改性磁性Fe3O4纳米颗粒载药量分别达到15.8%和9.3%。结论上述纳米药物载体在磷酸盐缓冲液中均表现出明显的缓释特性。[Objective] To assemble the medication 5-Fu for the treatment of cancer. [Methods] Fe3O4 nanoparticles as a kernel, 5-Fu, Fe3O4 and chitosan aqueous slurry were mixed by an appropriate proportion to produce the magnetic chitosan-5-Fu composed nanoparticles. Magnetes (Fe3O4) powder was treated by chemical method and transformed into nanometer size aqueous slurry. After Chitosan and Fe3O4 aqueous slurry were mixed and prepared by a optimized condition, the morphology and size of the magnetic chitosan-5-Fu composed nanoparticles were observed by TEM. Drug carried capacity and releasing properties in the phosphate buffer solution were investigated by UV-vis spectrum. [Results] Transmission electron microscope (TEM) shown the magnetes Fe3O4, the magnetic chitosan nanoparitcles and the magnetic chitosan 5-Fu shaped spherical form with a size rang of 8-12 nm, 12- 15 nm and 20-30 nm. The results tested by UV-Vis spectroscope to study the magnetic response displayed that magnetic chitosan nanoparticles had good magnetic properties. The observations shown that the magnetic chitosan- 5-Fu nanoparticles had spherical shape with a size range of 20-30 nm. The research results indicated that the carried capacity of 5-Fu-chitosan and magnetic chitosan-5-Fu nanoparticles were 15.8% and 9.3% respectively. [Conclusions] Drug releasing rate decreased with time lasting in above buffer.
