摘  要:目的探究慢性应激抑郁模型大鼠单胺类递质含量及递质间代谢关系,并从行为学及分子生物学角度探讨中药复方保神汤抗抑郁作用的机制及临床应用价值。方法采用孤养及慢性不可预见性中等强度应激的方法制成抑郁症大鼠模型,采用中药复方保神汤治疗,并与阳性药物(氟西丁)对照。通过观察动物的行为变化及测定大鼠海马及皮层单胺类递质以及其代谢产物的变化,观察该方对模型大鼠神经内分泌系统的影响。结果慢性应激与孤养可明显降低大鼠海马及皮层5-羟色胺(5-HT)、多巴胺(DA)含量,并使DA、去甲肾上腺素(NE)的代谢产物海马3,4二羟基苯乙酸(DOPAC)及内标物3,高香草酸(HVA)含量升高,5-羟吲哚乙酸(5-HIAA)/5-HT、DOPAC/DA比值升高,5-HT/NE降低,而中药高低剂量组均可逆转上述变化,疗效与氟西丁相当,尤以高剂量组明显。结论长期慢性应激造成的抑郁模型大鼠存在5-HT、DA等递质含量的不足及5-HT与NE的代谢失衡,保神汤可明显提高递质含量并纠正递质代谢的紊乱,使递质间关系趋于平衡,为临床防治抑郁症提供了新的方法与思路.Objective To explore the effect of Baoshentang on monoamine neurotransmitters and monoamine oxidase in brain of chronic stress induced-depression model rats for probing into it pathologically and molecular biologically anti-depress mechanism and clinical possibility of Baoshentang, a Chinese traditional herb prescription. Metheds Rat depression models were established by lonely raising and chronic unpredictable mild stress method. Depression model rats were administered by Baoshentang, with fluoxetine hydrochloride capsules as positive control.The changes of rat behavior were determined by open-field test and sucrose solution consumption test. The levels of monoamine neurotransmitters and their metabolites were detected by HLPC-ECD. Results Administration of Baoshentang and fluoxetine hydrochloride capsules improved depressive behavior. The levels of HE, 5-HT, DA and DOPAC in the hippocampus and cortex of chronic stress rats decreased. Baoshentang could reverse them partly (P〈0.05). Conclusions Chronic stress results in change of rat behavior and monoarnine neurotransmitters in brain. Administration of Baoshentang can prevent and recover those changes induced by chronic stress. Baoshentang can increase the levels of NE and 5-HT in the hippocampus and cortex of chronic stress rats. This may be its part mechanism of anti-depression.
