祛瘀生肌中药对糖尿病模型创面转化生长因子TGF-β1、TGF-β3水平的动态影响

摘　　要：目的:探讨祛瘀生肌中药对糖尿病大鼠愈创模型创面TGF-β1、TGF-β3水平的动态影响。方法:72只SD大鼠分别造成正常创面模型和糖尿病模型,随机分为生肌化瘀方组、化瘀方组和糖尿病创面生理盐水组。分别于不同干预时段(3d、7d、11d)取材,EnVision二步法免疫组化测定,并行图像扫描定量分析TGF-β1、TGF-β3水平变化。结果:生肌化瘀方及化瘀方组各时段(3d、7d、11d)TGF-β1及TGF-β3水平均高于糖尿病创面生理盐水组(P<0.05)。化瘀方TGF-β1均低于生肌化瘀方组(P<0.05),但TGF-β3各组比较无统计学差异(P>0.05)。结论:生肌化瘀方促进创面TGF-β1及TGF-β3分泌,有利于创面愈合,同时减少瘢痕形成。Objective To investigate the effects of Blood- Promotingand Stasis-Removing （祛瘀生肌） herbs on the synthesis of transforming growth factor （TGF-β 1 and TGF-β 3） in the granulation tissue of diabetic rat wound healing Methods Forty-eight male Sprague-Dawley（SD） diabetic rats were randomized into 4 groups（diabetic group, Shengji Huayu recipe-treated group, Huayu recipe-treated group and untreated group）. After the establishment of full thickness dermal wounding with the diameter of 2 cm on the back, the specimens were collected from the wound edge on the third, seventh and eleventh days after injury. The specimens were subjected to immunohistochemical methods and image analysis of tissue growth factor （TGF-β 1 and TGF-β 3）. Results The expression of TGF-β 1 at tile third, seventh and eleventh days in Shengji Huayu recipe-treated group and Huayu recipe-treated group granulation tissue were higher than that of the diabetic group, but the expression of TGF-β 1 of Huayu recipe-treated group was lower than that of Shengji Huayu recipetreated group; there was no statistically difference of TGF-β 3 between Shengii Huayu recipe-treated group and Huayu recipe-treated group. Conclusion Shengji Huayu recipe-treated promotes TGF-β 1 and TGF-β 3 expression to acceler- ate healing, and avoids forming cicatricle at the same time.
