摘 要:目的探讨复方丹参方中使药冰片对君药丹参在新西兰大白兔体内药代动力学过程的影响。方法丹参组、丹参-冰片配伍组水煎液灌胃给药,液-液萃取法提取血浆中的丹参素,以对羟基苯甲酸为内标,采用离子阱质谱多级反应模式(MRM)测定兔血浆和脑脊液中丹参素的质量浓度,DAS数据处理软件计算药动学参数。结果丹参-冰片配伍给药可使丹参中丹参素的药动学参数t1/2αt、1/2β、kα、AUC(0-∞)较单独给药时明显增大,丹参素的脑血比提高。结论复方丹参方中,使药冰片可提高君药丹参中丹参素的生物利用度,减慢代谢,促进血脑屏障对丹参素的通透性。该研究可为“君-使”对药体内作用机制的研究提供思路。Objective To study the pharmacokinetic effects of borneol (Shi herb) on Salvia Miltiorrhiza (Jun herb) in Fufang Danshen prescription. Methods Water extracts of Salvia Miltiorrhiza and Salvia Miltiorrhizaborneol were orally administered to New Zealand rabbits. Plasma samples were pretreated by liquid-liquid method, and 4-hydroxybenzoic acid was used as the internal standard. The multi-reaction mode supplied by the Chemistation software was applied to determine the concentration of Danshensu in plasma and cerebrospinal fluid (CSF). The data were processed by DAS to calculate the pharmacokinetic parameters. Results Shi herb significantly increased the parameters of t1/2α,t1/2β,kα,AUC(0-∞) and CSF-plasma ratio. Conclusion In Fufang Danshen prescription, Shi herb could increase the bioavailabilty and enhance the permeability of Jun herb, but slow down metabolism. This work can provide the thoughts for substantial foundation study of "Jun-Shi" pairing herbs.
