摘 要:目的探讨缺氧诱导因子-1α在支气管哮喘大鼠模型支气管肺组织中的表达及意义。方法采用卵蛋白致敏诱导建立支气管哮喘大鼠模型,采用逆转录半定量PCR及免疫印迹技术检测正常对照组、阴性对照组、哮喘组和地塞米松组支气管肺组织中缺氧诱导因子-1α基因和蛋白表达的水平。结果与正常对照组比较,哮喘组缺氧诱导因子-1α基因和蛋白表达水平均显著增加,差异有显著性(P<0.05,P<0.05);与正常对照组比较,阴性对照组缺氧诱导因子-1α基因和蛋白表达水平没有明显改变,差异均无显著性(P>0.05,P>0.05);与哮喘组比较,地塞米松组中的缺氧诱导因子-1α基因和蛋白表达水平均显著性降低,差异有显著性(P<0.05,P<0.05),但与正常对照组比较,差异均无显著性(P>0.05,P>0.05)。结论缺氧诱导因子-1α可能参与支气管哮喘的发病机制且其表达的水平受地塞米松调节。Abstract Objective To investigate the expression of hypoxia inducible factor-1α ( HIF-1α) in bronchopulmonary tissue in asthmatic rat model and its significance. Methods Thirty-two adult Wister rats were randomly divided into normal control group, negative control group, asthma group and asthma group treated with dexamethasone (DXM) groups. Asthma rat model was established by sensitization and stimulation with ovalbumin (OVA) conjuncted with AI(OH)3. RT-PCR and Western Blot were used to evaluate the gene and protein expression of HIF-1α in bronchopulmonary tissue in these groups. Results Compared gene and protein levels increased significantly in asthma to the control group, the expression of HIF-1 ct both in group ( P 〈 0. 05, P 〈 0. 05 ). However, there was no significant difference between the control and negative group ( P 〉 0. 05, P 〉 0. 05, ). The expression of HIF-1α in asthma group treated with DXM was significantly reduced when compare to asthma group ( P 〈 0. 05, P 〈 0. 05 ). But no significant difference of expression levels was detected between asthma group treated with DXM and control group(P 〉 0. 05 ,P 〉 0. 05 ). Conclusion HIF-1α may be involved in asthmatic pathogenesis, and its expression level may be regulated by DXMI
