芍药苷对佐剂性关节炎大鼠腹腔巨噬细胞吞噬功能及其产生细胞因子的影响

摘　　要：目的研究芍药苷(PF)对佐剂性关节炎(AA)大鼠腹腔巨噬细胞(PMΦ)吞噬功能及其产生细胞因子的影响。方法采用弗氏完全佐剂(FCA)诱导AA大鼠模型,无菌取PMΦ,吞噬功能采用中性红法测定,PMΦ培养上清液中IL-1活性的测定采用小鼠胸腺细胞增殖法,TNF-α和PGE2含量的测定采用放射免疫测定法。结果PF能抑制关节肿胀,降低AA大鼠PMΦ过强的吞噬功能和产生IL-1、TNF-α和PGE2的水平。结论PF对AA大鼠的抗炎和免疫抑制作用与其降低AA大鼠PMΦ过强的吞噬功能和产生IL-1、TNF-α和PGE2的水平有关。Objective To study the effect of paeoniflorin （PF） on peritoneal macrophage from rats of adjuvant-induced arthritis（AA）. Methods Freund＇s complete adjuvant was used to induce AA in rats. The phagocytotic activity of peritoneal macrophage in AA rats was determined by natural red method, and interleukin-1 （ IL-1 ） activity in peritoneal macrophage （TNF-α） and prostagland supernatant was measured by thymocyte proliferation assay. Tumor necrosis factor alpha in E2 （PGE2 ）produced by peritoneal macrophage were determined by radioimmunoassay. Results PF was shown to relieve swelling of joint to some extent. Meanwhile, PF also suppressed the over-production of IL-1 ,TNF-α and PGE2. Conclusion PF has antiinflammatory effect,which its mechanism may be related to the modification of the abnormal immunological function of peritoneal macrophage from AA rats.
