摘 要:目的观察细胞外ATP对臂丛神经损伤后失神经骨骼肌和前角运动神经元的保护作用。方法健康雌性Wistar大鼠60只,随机分为实验组和对照组,制作大鼠左侧臂丛神经根性切断模型。术后每日实验组腹腔注射ATP(2 mg/kg)0.4 mL,对照组注射生理盐水0.4 mL。术后2、4、6周,用免疫组织化学方法检测臂丛神经根性切断后脊髓C_5-T_1的神经营养素-3(NT-3)的表达活性。术后4、6周取肱二头肌用透射电镜观察肌肉超微结构的变化。结果与对照组相比,各实验组术后2、4、6周脊髓NT-3的表达增多,差异有统计学意义(P<0.05)。肱二头肌超微结构显示:实验组线粒体肿胀及肌质网扩张程度轻、间质胶原纤维和成纤维细胞少、肌丝肌节排列整齐无明显紊乱,线粒体嵴较多,形状相对正常,线粒体周围糖原颗粒增多。结论细胞外ATP对臂丛神经切断后的脊髓运动神经元和失神经骨骼肌有保护作用。Objective To observe the influence of ATP protection after brachial plexus injuries. Methods A total of 80 female Wistar rats, weighting 280 - 300 g, were randomly divided into ATP and control groups. The right C5 -TI nerve roots were transected and then the intraperitoneal injection of 4ml of ATP or normal saline was given immediately and once daily to the rats, respectively. The rats were sacrificed on postoperative days 14, 28 and 42 respectively. The C5-T1 segments of the spinal cord were harvested. NT-3 activity was measured by enzymo-histochemistry method. Four weeks and 6 weeks postoperatively, ultrastructure of the denervated skeletal muscles was observed. Results Compared to the control group, the expressions of NT-3 was increased in the treated groups with ATP injection ( P 〈 0. 05 ). There was less degenerated nucleus, minor mitochondriaedema and sarcoplasmic reticulum enlargement, less interstitial fibroblast and collagenfibers, better oriented myomere and myofilament were seen in the experimental groups. Conclusion Extracellular ATP have neurotrophic effect on neurons survial and could improve the expression of NT-3 in the tissues of the spinal cord, ATP can protect the ultrastructure of denervated skeletal muscle.
