猪捷申病毒中国株3ABC基因的克隆及序列分析

摘　　要：根据已报道的Swine/CH/IMH/03基因序列设计1对引物,从已分离的中国株捷申病毒中提取总RNA,经RT-PCR扩增得到3ABC基因.并将该基因克隆到pMD18-T载体中,经序列测序,并与GenBank上的14种毒株序列进行比对.结果表明:猪捷申病株中国株(PTV-3ABC)与F65毒株、PTV-1毒株、Swine/CH/IMH/03毒株和Talfan毒株的同源性比较高,分别为96.5%,99.5%和99.4%分别为99.4%,氨基酸同源性分别为95.6%,98.8%,98.4%和98.8%.系统发育树表明,此中国株捷申病毒属于PTV-1型.Based on the reported sequences of Swine/CH/IMH/03, a pair of primers to amplify the 3ABC gene was designed. The total RNA was extracted from China strain of porcine teschen virus,and the 3ABC gene was amplified by RT- PCR. The products of PCR were cloned into pMD- 18T vector, sequenced and then the sequences were aligned with 14 sequences from GenBank. The results showed that its homology with F65, PTV- 1, Swine/CH/IMH/03 and Talfan strain was very high, showing 96.5 %- 99.4 % identity in nucleotide sequence and 95.6 % -98.8% identity in deduced amion acid sequence. According to the phylogenetic tree,the China strain of teschen virus should belong to PTV- I.
