更年甘露饮加小剂量雌激素对成骨细胞增殖及BMP-2表达的影响

摘　　要：目的通过观察复方中药更年甘露饮加小剂量雌激素对成骨细胞增殖及骨形成蛋白(BMP)-2的影响,探讨一种治疗绝经期骨质疏松的新方法。方法分别用模型血清、更年甘露饮血清、更年甘露饮血清加1/8、1/4、1/2最佳量雌激素及模型血清加最佳量雌激素用于第3代成骨细胞,用MTT法检测成骨细胞增殖,用ELISA及免疫组化法检测成骨细胞BMP-2的表达。结果更年甘露饮加入1/8、1/2、1/4最佳量雌激素时均可提高对成骨细胞的作用,加入1/4与1/2最佳量雌激素时作用更明显,增殖较最佳量雌激素组明显,BMP-2表达与最佳量雌激素组相近。结论更年甘露饮加小剂量雌激素可明显提高成骨细胞增殖及BMP-2蛋白表达,此方法可能作为一种治疗绝经后骨质疏松的治疗方法。Objective To investigate the effects of Gengnianganluyin plus low-dose estrogen on proliferation and BMP-2 expression of rat osteoblast and to approach a new treatment to prevent menopausal cardiovascular disease. Methods Osteoblasts were divided into mod- el serum group, Gengnianganluyin group,Gengnianganluyin plus 1/8,1/4,1/2 optimal estrogen group and model serum plus optimal estrogen group. Cell proliferation and BMP-2 protein expression were determined by MTY and ELISA respectively. Results Compared with Gengnianganluyin group,Gengnianganluyin plus 1/8 optimal estrogen was effective and plus 1/4 as well as 1/2 estrogen optimal estrogen were more effective,which got close to optimal estrogen. Conclusions Gengnianganluyin plus low-dose estrogen can improve osteoblast proliferation and BMP-2 expression as well as normal-dose estrogen, which could be a new treatment to prevent postmenopausal osteoporosis.
