摘 要:目的:建立新化合物1,3-二氧咖啡酰甘油酯HPLC测定方法,测定菠萝叶中1,3-二氧咖啡酰甘油酯的含量。方法:采用Kromasil C18色谱柱(4.6 mm×150 mm,5μm)进行分离,流动相磷酸溶液(0.1%)-乙腈(75∶25),流速1.0 mL.min-1,柱温30℃。结果:1,3-二氧咖啡酰甘油酯与其他组分分离效果较好,在0.051 6μg^0.516μg具有良好的线性(r=0.999 9),平均回收率为97.1%(RSD 1.3%)。经测定3批菠萝叶中1,3-二氧咖啡酰甘油酯的平均质量分数分别为0.033%,0.034%,0.031%。结论:该分析方法具有较好的分离效果以及很高的回收率和重复性,可以用于菠萝叶中1,3-二氧咖啡酰甘油酯的含量分析及质量控制。Objective: To establish a HPLC method for a new compound 1, 3-O-dicaffeoylglycerides determination. Method:The separation was performed in a Kromasil C18 column (4.6 mm × 150 mm, 5 μm) with a mobile phase of 0. 1% H3PO4 : Acetontrile = 75:25 (v/v). The flow rate was 1.0 ml · min^-1 and the temperature of column was 30 ℃. Result: A satisfactory separation between 1, 3-O-dicaffeoylglycerides and impurity was obtained. The calibration curve was linear over the concentration range from 0.051 6μg to 0. 516 μg. r =0. 999 9. The average recoveries was 97. 1% (RSD 1.3% ). The content of 1,3-O-dicaffeoylglycerides in pineapple leaves from three different batches were 0. 033%, 0. 034% and 0. 031% respectively. Conclusion: The method has good selectivity, high reeoverv and reproducibility, and can be used for the analysis of 1, 3-O-dicaffeoylglycerides in pineapple leaves and their quality control.
