摘  要:背景与目的:观察急性镉中毒大鼠肾皮质中血红素氧化酶-1(heme oxygenase-1,HO-1)表达的变化。材料与方法:健康雄性SD成年大鼠24只,随机分为4组,3个染镉组连续5d分别腹腔注射0.4、0.8、1.6mg/kg氯化镉(CdCl2);对照组连续5d腹腔注射生理盐水。给药结束后5d取双肾皮质分别进行RT-PCR及免疫组织化学分析,观察HO-1mRNA及蛋白表达的变化。结果:对照组观察到HO-1的生理性表达。在mRNA水平,0.4mg/kg组未见表达增加,0.8及1.6mg/kg组较对照组明显升高(P<0.05)。mRNA与染镉剂量呈剂量-效应关系(r=0.97,P<0.05)。免疫组化结果显示,肾皮质HO-1蛋白表达的变化较mRNA敏感。在蛋白水平,3个试验组与对照组相比,差异均有统计学意义(P<0.05)。结论:镉的急性毒性作用可诱导肾脏表达HO-1增高,其表达可能具有保护作用。BACKGROUND & AIM: To observe the changes of heme oxygenase-1 (HO-1) expressed in renal cortex of rats with acute cadmium exposure. MATERIALS AND METHODS: 24 male SD rats were divided randomly into 4 groups. There were a control group and the three experimental groups received daily intraperitoneal injections of CdCl2 (0.4, 0.8, 1.6 mg/kg) for five days. 0.9% NaCl substituted for CdCl2 in control. The specimens of kidney were prepared to measure the expression of HO-1 by RT-PCR and immunohistochemistry technique on the fifth day after injection. RESULTS: The physical expression of HO-1 was detected in control group. The levels of expressed mRNA in 0.8 and 1.6 mg/kg groups increased dramatically (P 〈 0.05) compared with the control while the mRNA concentration of 0.4 mg/kg group was equal to the control. A dose-effect relationship between the cadmium exposure and expression of HO-1 mRNA(r = 0.97, P 〈 0.05) was also found. Analysis of immunohistochemistry showed that changes of protein expression was more sensitive than mRNA in all three groups compared with the control(P〈0.05) . CONCLUSION: The acute exposure to cadmium could induce HO-1 expression in renal cortex of rats and this might provide potential protection.
