摘 要:目的探讨C反应蛋白(CRP)及其单体(mCRP)对人脐静脉内皮细胞晚期糖基化终产物受体(RAGE)蛋白表达及mRNA的影响。方法将人脐静脉内皮细胞用不同浓度(5、25、50、100μg/mL)的CRP孵育24h及同一浓度(50μg/mL)CRP不同时间孵育0、6、12、24、48h,采用流式细胞仪及RT-PCR方法检测RAGE蛋白及mRNA的表达水平。分析50μg/mLCRP和mCRP孵育0、6、24h后对RAGE蛋白表达的影响。结果(1)与对照组比较,5μg/mL的CRP培养内皮细胞24h显著增高RAGE蛋白的表达(175.2%±8.1%,P<0.05),50μg/mL时达到顶峰(527.3%±5.8%,P<0.01);50μg/mLCRP孵育不同时间,RAGE蛋白的表达从6h明显增高(179.8%±29.1%,P<0.05)。(2)与对照组比较,5μg/mLCRP即可升高RAGE的mRNA表达(135.4%±11.1%,P<0.05),并在50μg/mL时达到峰值(186.6%±29.0%,P<0.05);6h时可以检测到RAGE mRNA的升高(134.3%±22.8%,P<0.05),在12h为最高(214.2%±33.1%,P<0.05)。(3)mCRP也会升高RAGE蛋白表达,与CRP组没有差别(P>0.05)。结论C反应蛋白以时间及剂量依赖的方式促进内皮细胞RAGE蛋白及mRNA的表达。CRP与其单体(mCRP)作用相似。Objective To investigate the effect of C reactive protein on receptor of advanced glycation endproducts protein (RAGE) and mRNA expression in human endothelial cells. Methods Human saphenous vein endothelial cells (HSVECs) were incubated with CRP. RAGE protein levels were determined by flow cytometry, and RAGE mRNA levels were assessed by RT-PCR. Results (1)Compared with control group (100%), CRP caused a significant increase in RAGE protein expression at a dose as low as 5 μg/mL ( 175.2 ±8. 1%, P〈 0. 05) and reached peak value at 50 μg/mL (527.3 ± 5.8%, P〈0.01). RAGE protein expression was elevated (179. 8 ±29.1%, P〈0. 05) as early 6 hours after CRP incubation. (2)At the rnRNA level, RAGE gene expression was increased by CRP in a dose dependent manner as well. RAGE mRNA reached its peak value at 12 hours (214.2 ± 33. 1%, P〈0.05). (3)Effect of monomeric CRP on RAGE protein expression were the same as that of native pentameric CRP (P〈0. 05). Conclusion C-reactive protein upregulates RAGE protein and mRNA expression in human endothelial cells in a dose and time dependent manner.
