摘 要:目的观察小鼠皮下接种Ⅰ型单纯疱疹病毒(HSV-1)产生的神经痛,建立急性期疱疹疼痛模型。方法120只Balb/c雌性小鼠,随机分为2组:对照组和实验组,对照组10只,实验组110只。对照组于左后肢胫部皮下接种灭活的HSV-1;实验组于同部位皮下接种HSV-1,分别于接种前、接种后第1、2、3、4、5、6、7、8、10、12天各随机取10只小鼠,于当日9:00 am观察皮肤脱毛区变化,并测定其热痛潜伏期,4:00 pm测定同批小鼠机械痛觉反应评分。测定完毕后处死小鼠,采用聚合酶链反应检测背根神经节中HSV-1 DNA。结果实验组接种后第5天小鼠均出现皮损,第7天均达高峰,以后逐渐恢复;与接种前比较,接种后第5天出现机械痛觉反应评分明显增高,直至第12天仍较高(P<0.05);而热痛潜伏期两组均无明显变化(P>0.05);接种后第2~12天在背根神经节中均可观察到HSV-1 DNA,而对照组则无此变化。结论小鼠左后肢胫部皮下接种HSV-1后产生了相应的皮肤损害和机械痛敏,此法成功建立了急性期疱疹疼痛模型。Objective To establish a mouse model for acute herpetic pain induced by subcutaneous inoculation of herpes simplex virus type-1 (HSV-1) in the hind leg of the mice. Methods One hundred and twenty Balb/c mice were randomly allocated into 2 groups: experiment group (n = 110) and control group (n = 10). In the experiment group the mice were inoculated with HSV-1 subcutaneously at the left shin, while in control group the mice were inoculated with devitalized HSV-1. The latency for left hind paw to withdraw from heat source and the withdrawal threshold of left hind paw to yon Frey hair stimulation were measured and recorded the day before inoculation and on day 1, 2, 3, 4, 5, 6, 7, 8, 10 and 12 after inoculation. The skin of the left shin where HSV-1 was inoculated was examined and HSV-1 DNA in the dorsal root ganglions was detected.Results The skin lesion appeared on day 5 after inoculation and got worse on day 7 then gradually disappeared. Hyperalgesia to mechanical stimuli started on day 5 after inoculation and persisted until day 12, but thermal hyperalgesia was not observed. HSV-1 DNA was detected in the dorsal root ganglions from day 2-12. Conclusion After being inoculated with HSV-1 the mice developed skin lesion and mechanical hyperalgesia. This can be a useful model for studying the mechanism of herpetic pain.
