摘 要:目的研究在高糖条件下人血管平滑肌细胞丝裂原蛋白激酶(MAPK)活性、c-fos表达与糖尿病慢性血管并发症的发病机制的关系。方法新生儿脐动脉原代培养获得人血管平滑肌细胞。高糖为刺激因素,PD98059为MAPK特异性抑制剂,甘露醇作渗透压对照。同位素示踪法测定MAPK活性,免疫细胞化学方法检测c-fos表达。结果高糖条件培养下人血管平滑肌细胞内MAPK活性升高(77358±1822)、与对照组(47830±1946)相比差异有统计学意义(P<0.05)。高糖条件培养下人血管平滑肌细胞c-fos表达增加(0.314±0.058),与对照组(0.245±0.049)相比差异有统计学意义(P<0.05)。PD98059可阻止高糖诱导的c-fos表达。结论高糖可增加人血管平滑肌细胞MAPK活性及c-fos的表达,MAPK活性的增高与c-fos表达呈正相关。Objective To investigate the activity of MAPK and the expression of c-fos in human vascular smooth muscle cells cultured in high glucose and the mechanism of chronic vascular complication of type 2 diabetes. Methods The human smooth muscle cells were obtained by primary cell culture. They were stimulated by high glucose. PD98059 was a special inhibitor of MAPK. The MAPK activity was measured by liquid scintillation counting. The expression of c-fos was measured by immunochemistry. Results Compared with the control group (47830±1946), the high glucose increased the activity of MAPK (77358±1822) (P〈0. 05). Compared with the control group (0. 245±0. 049), the expression of c-los increased (0. 314 ± 0. 058) (P〈0.05). PD98059 inhibited the activity of MAPK. Conclusions The activity of MAPK and the expression of c-fos are increased when cells were cultured in high glucose. The activity of MAPK affects the expression of c-fos.
