摘  要:目的探讨神经于细胞(NSCs)移植到慢性海人酸(KA)致癫痫鼠海马CA3区后对其认知功能的影响。方法采用KA脑室注射法制作慢性癫痫鼠模型,将原代培养的增强绿色荧光蛋白(EGFP)标记的NSCs移植到慢性癫痫鼠的海马CA3区,所有大鼠在移植组移植后12周行Morris水迷宫试验,然后移植组大鼠取脑冰冻切片,在倒置荧光显微镜下直接观察移植细胞的存活和迁移,采用免疫荧光染色法观察移植细胞分化情况。结果癫痫鼠组大鼠在目标区域逗留时间明显短于正常大鼠组,而逃避潜伏期明显长于正常大鼠组(P<0.01);移植组大鼠的逃避潜伏期比癫痫组和假手术组大鼠明显缩短,在目标区域逗留时间显著延长(P<0.01)。且移植后12周,NSCs大量存活(65045.00±881.72),迁移到齿状回和海马各区并分化为神经元和神经胶质细胞。结论癫痫能够损害大鼠的认知功能。将NSCs移植于KA致慢性癫痫鼠海马CA3区后,NSCs不仅能够长期存活、迁移到齿状回和海马各区并分化成神经元和神经胶质细胞,而且能够明显改善癫痫鼠的认知能力。Objective To evaluate the influence on the cognitive function of adult chronic epileptic rats after transplanting neural stem cells (NSCs) to their hippoeampal CA3 region. Methods The model of adult chronic epileptic rats was created by intracerebroventicular injection of kainic acid (KA). Neural stem cells were primarily cultured and labeled with the enhanced green fluorescent protein (EGFP) and transplanted into hippoearnpal CA3 of adult ehronie epileptic rats treated with KA. Morris water maze tests were performed 12 weeks after transplantation of NSCs. Survival and migration of the NSCs were detected directly under a phase contrast fluorescent microscope and the differentiation of the NSCs was determined by immunoeytoehemical fluorescent staining. Results After KA treatment, the time of the rat' escape latency was longer and the time of staying at goal region was shorter than normal group rats (P 〈 0.01 ). At 12th week after transplantation of NSCs ,the time of the rats'escape latency was shorter and the time of staining at goal region was longer than the KA-treated and sham-operation groups (P 〈 0.01 ). There was no difference between sham-operation group and KA-treated group. And at 12th week after transplantation of NSCs, large number of the grafted cells existed ( 65 045.00 ±881.72) and these ceils migrated to dentate gyms and all area of hippoeampi and differentiated into neurons and astroeytes. Conclusion Epilepsy causes cognitive damage of rats. The NSCs after transplantation into hippocampal CA3 of adult chronic epileptic rats not only can migrate to dentate gyms and all area of hippocampus and survive well for long time,but also can differentiate into neurons and astroeytes and ameliorate the cognitive function of rats.
