摘  要:为了便于全面了解目前细胞内电处理实验研究的方法和具体实施方案,综述了国内外用于细胞内电处理实验研究的ns脉冲发生器。从电容器充放电和传输线理论2方面介绍了这些发生器产生ns级、高强脉冲电场的工作原理;分析了用于细胞内电处理的ns脉冲发生器性能特点、发生器的电路结构、元器件选择和技术指标等。实验结果表明,这些发生器产生的ns脉冲电场能诱导细胞内电处理效应。对今后的研究工作进行了讨论和展望。Unlike the principle of eleetroporation for cellular outer membrane, the principle of intracellular electromanipulation (IEM) is that nanosecond pulsed electric fields (nsPEF) with amplitude above 10kV/cm can affect the subeellular structures without leading to the formation of large bores in the outer membrane. IEM effects, such as release of calcium ion from intracellular organdies, enhancement of gene expression and inducement of cell apoptosis and so on, have shown tremendous application potentials in tumor treatment and other applications. A generator plays an important role in experiment research on IEM, which is capable of producing pulsed electric field with duration in the order of nanoseconds and strength above 10kV/cm. In order to understand experiment research on IEM more comprehensively, this paper aimed at presenting a review on nanosecond pulse generators for the study of IEM. These pulse generators produced nsPEF in given treatment chamber, and IEM was studied from the point of view of experiment phenomena. As to the storage type, the generators are capacitive storage. According to the principle of producing nanosecond pulse, the generators can be categorized as three types. The first is charging a capacitor and then discharging it on the load, the other is transmission line theory that transmission process of electromagnetic wave is utilized to produce pulse with given pulse duration. Then, the circuit design, component selection, typical waveforms, advantages and disadvantages of these generators are described briefly. Moreover, some valuable issues for future exploration in this area are also indicated and discussed.
