摘  要:目的探讨丹皮多糖的相对分子质量与其生物活性的关系。方法运用正交设计方法,以酸浓度、水解温度、水解时间为因素,分别设置4个水平,对PSM2b-A进行降解和结构改造,经快速色谱纯化系统分离,选择相对分子质量适宜的片段,以抑制体外非酶糖化反应(NEG)为模型,进行活性检测。结果当H2SO4浓度为0.5mol.L-1时,50℃保温6h和80℃保温2h降解得到的2个片段F1和F2都有明显抑制体外NEG反应的作用,抑制率分别达到35.26%和27.56%。且F1的抑制率高于PSM2b和阳性对照氨基胍。结论相对分子质量与PSM2b-A抑制体外NEG活性关系明显,降低PSM2b-A的相对分子质量可以提高其抑制NEG反应的活性。OBJECTIVE To study relative the relative molecular mass of Mudan cortex(PSM2b-A ) on the inhibitory effects of nonenzymatic glycosylation(NEG) of protein in vitro. METHODS PSM2b-A was degraded by acid hydrolysis through orthogonal experimental design. The density of acid, the time and temperature of the reaction were the three factors which had influence on the results of the hydrolysis of PSM2b-A. The degradation products were separated by AKTA purifier,suitable fractions through non-enzymatic glycosylation of protein in vitro were screened.RESULTS Under the condition of 0.5 mol·L^-1 H2SO4 at 50℃ for 6h and 80℃ for 2h, two fractions named F1 and F2 inhibited NEG in vitro and the inhibiting rates reached 27. 56% and 35. 26%. The inhibiting rate of F1 even exceeded the PSM2b and positive control. CONCLUSION The relative molecular mass has a significant relationship with the activity of PSM2b-A in inhibiting the NEG in vitro. And the decrease of the relative molecular mass of PSM2b improves the activity in inhibiting the NEG in vitro.
