摘  要:目的对肾癌细胞株中XIAP(X-linked inhibitor of apoptosis)的表达进行基因水平和蛋白水平分析,了解XIAP表达与肾癌进展的关系。方法采用相对定量实时荧光PCR和Western blot方法,分析9株来源于不同分期和组织学类型的肾癌细胞株中XIAP在mRNA和蛋白质水平的表达。结果pT3a~pT3b肾癌细胞株、pT2肾癌细胞株和正常对照组的XIAP相对mRNA表达水平(即XIAP/GAPDH比值)分别为13.25×10^-3±8.22×10^-4、8.37×10^-3±1.38×10^-4和3.93×10^-3±5.88×10^-4,3组间差异有统计学意义(P〈0.01)。Western blot方法显示pT3肾癌细胞株XIAP表达量明显高于pT2肾癌细胞株和正常对照组。结论肾癌细胞中XIAP在mRNA和蛋白水平呈肿瘤进展依赖性增高的特点,为肿瘤细胞抗凋亡提供了条件,可能与肾癌的进展有关。Objective To explore the relevance of anti-apoptotic XIAP for tumor progression in renal cell carcinomas (RCC), by the analysis of mRNA and protein expression of XIAP in RCC cell lines derived from primary RCC. Methods mRNA and protein expression of XIAP were analyzed by relative quantitative real-time PCR and Western blot respectively, in 9 of RCC cell lines with different pathological stages and histological types. Results XIAP mRNA expression was found in all RCC cell lines. The relative mRNA expression levels (XIAP/GAPDH ratios) were detected as 13.25 × 10^-3 ±8. 22 )〈10^-4 in pT3a/pT3b , 8.37 × 10^-3 ± 1. 38 × 10^-4 in pT2 and 3. 93 × 10^-3 ±5. 88 ×10^-4 in normal kidney tissue by relative quantitative real-time PCR, Importantly, the XIAP mRNA expression levels were found significant increased in RCC cell lines from earlier (pT2) to advanced (pT3a, pT3b) tumor stage. Western blot analysis confirmed the tumor stage-dependent increase of XIAP expression on the protein level. Conclusions Anti-apoptotic XIAP expression gradually increases during progression of RCC, probably contributing to the marked apoptosis-resistanee of RCC, thereby prompting tumor progression,
