摘 要:目的 探讨冲突性应激对大鼠额叶代谢物N-乙酰天冬氨酸(NAA)水平的影响。方法21只SD雄性大鼠,随机分为3组:正常对照组,4周应激组,4周应激用药组。采用酶联免疫吸附试验检测血清中皮质酮的含量,高效液相色谱检测额叶NAA浓度。结果4周应激组大鼠皮质酮的水平[(161.02±17.86)ng/ml]比其对照组[(89.23±12.34)ng/ml]的高(P〈0.01),4周应激用药组大鼠皮质酮的水平[(83.5±9.33)ng/ml]已经基本上恢复到对照组水平(P〉0.05),而远远低于4周应激组(P〈0.01)。与正常对照组[(6.23±0.61)μmol/g]相比较,4周应激组NAA浓度[(5.17±0.54)μmol/g]下降,差异具有显著性(P〈0.05);与应激用药组[(6.43±0.83)μmol/g]比较,4周应激组代谢物浓度显著下降(P〈0.01)。用药组和正常对照组NAA浓度的差异无显著性。结论 冲突性应激可以诱发皮质酮水平的升高,脑代谢物N-乙酰天冬氨酰含量下降,帕罗西汀一定程度上可以改善这种情况。Objective To investigate changes of N-acetylaspartate(NAA) in frontal cortex of rats in response of chronic conflict-related stress. Methods 21 male SD rats were randomly divided into 3 groups: con- trol, 4-week consistent stress, 4-week consistent stress with antidepressant drug treatment (paroxetine HC1). After the last sessions,plasma corticosterone with enzyme-linked immunosorbent assays and NAA levels with high performance liquid chromatography were determined. Results Corticosterone levels for the 4-week stress rats were significantly higher than that for control ( P〈0.01 ) and stressed rats with drug treatment ( P〈0.01 ). There was not a significant difference between non-stressed animals and stressed with drug treatment ( P〉 0.05 ). The concentration of NAA in the frontal cortex of 4-week stress group was less than the concentration of the drug group ( P 〈0.01 ) and the control ( P〈 0.05). Conclusion The concentration of NAA in frontal cortex of rats decreased following the conflict stress. All these changes could be improved by the paroxetine.
