摘 要:目的分析探讨人载脂蛋白 CⅢ(ApoCⅢ)基因多态性与脂代谢异常和冠心病的相关性。方法以400名健康献血者和360例冠心病患者为研究对象,经静脉采血后进行血生化检测和提取全血 DNA,通过荧光实时定量 PCR(FQ-PCR),同时分析载脂蛋白 CⅢ-625和-455位点变异与冠心病、血脂的关系。结果冠心病组载脂蛋白 CⅢ-455C 纯合子基因型(101例,70.1%),明显高于对照组(85例,53.1%),-625del-455c 基因型(49.3%)显著高于对照组(41例,25.6%)。单纯高脂血症组和高脂血症并发冠心病组,载脂蛋白 CⅢ-455C 基因表达比较高。单纯冠心病组的载脂蛋白 CⅢ-455C 基因表达与正常组间差异无统计学意义。结论载脂蛋白 CⅢ-455C 基因与冠心病风险呈正-相关,-455C 纯合子基因型携带者具有冠心病的高风险。载脂蛋白 CⅢ-625(del)与-455(C)有协同增加冠心病风险的作用。Objective To explore the relation between apolipoprotein C Ⅲ (ApoC Ⅲ ) gene polymorphism and coronary artery disease (CAD). Methods Peripheral blood samples were collected from 400 healthy blood donors and 360 CAD patients. Total cholesterol (TC) , triglycerides (TG), high-density lipoproteins cholesterol (HDL-C), low-density lipoproteins cholesterol (LDL-C), and ApoC Ⅲ were detected. Fluorescence quantitative PCR was used to detect the ApoC Ⅲ ) gene polymorphisms. Genomic DNA was extracted from whole blood samples. We study the relation between variants of ApoC Ⅲ-625 and -455 and CAD by FQ-PCR. Then we explore the relation between variants of ApoC Ⅲ-625 and -455 and the concentration of lipid. All objects were measured about Total Cholesterol(TC) , Triglycerides (TG), high-density lipoproteins cholesterol ( HDL-C ), low-density lipoproteins cholesterol ( LDL-C ) and ApoC Ⅲ. Results The ApoCⅢ-455C homozygote rate of the CAD patients was 70.1%, significantly higher than that of the healthy persons (53. 1%). The ApoC Ⅲ ,-625del-455c phenotype rate of the CAD patients was 49. 3%, significantly higher than that of the healthy persons. The ApoC Ⅲ-455C expression rates of the patients with hyperlipidemia and those with hyperlipidemia combined with CAD were high. There was no significant difference in the expression of ApoC Ⅲ-455c between the CAD patients and the normal persons. Conclusion The gene polymorphisms of ApoC Ⅲ-455 increase the risk of CAD. ApoC Ⅲ-455C gene is positively related to the risk of CAD. The homozygotes of the ApoC Ⅲ-455C gene have high risk of CAD. AooCⅢ-455C in coooeration with -625( del) will increase the risk of CAD.
