摘  要:目的在体观察α-晶体蛋白对LongEvens大鼠视神经损伤后视网膜神经节细胞(RGCs)存活的保护作用。方法于视神经损伤前7d经上丘及外侧膝状体逆行标记RGCs。成年LongEvens大鼠35只,其中10只用于提取α-晶体蛋白,另外25只分成5组,分别为正常对照组和伤后1,2,3,4周组,每组5只。右眼经玻璃体腔注射浓度为1×10^-5g/L的α-晶体蛋白5μl,左眼注射牛血清白蛋白5μl,分别于伤后1,2,3,4周对实验大鼠行RGCs计数。结果视神经夹伤后1周RGCs数明显下降,牛血清白蛋白组下降为正常值的33%;视神经夹伤后2周下降为正常值的4%,3周时下降为正常值的3%,4周时下降为正常值的1%。玻璃体腔注射α-晶体蛋白组在视神经夹伤后1周RGCs数下降为正常值的51%;2周时RGCs数下降为正常值的11%;3周时RGCs数下降为正常值的7%,4周时下降为正常值的3%,均显著高于对照组(P〈0.01)。结论视神经损伤后玻璃体腔注射可溶性α-晶体蛋白能够延缓RGCs死亡,具有一定的保护作用。Objective To investigate the protective effect of intraocular injection with α-crystallin in vivo on the survival of retinal ganglion cells (RGCs) after optic nerve injury. Methods Among 35 Long Evans rats, 10 were used forα-crystalline extractionand the other 25 were dived equally intocontrol group, 1 week, 2 weeks, 3 weeks and 4 weeks groups after lesion. RGCs were labeled by fluorogold retrogressively through superior colliculus and lateral geniculate body seven days before optic nerve injury. The vitreous of the right eyes were injected with 5 μl α-erystallin ( 1 ×10^-5g/L) and that of the left eyes with 5 μl bovine serum albumin (BSA). The RGCs were counted 1,2,3,4 weeks after lesion. Results The number of RGCs began to decrease 1 week after lesion. In the BSA group, the number of RGCs decreased to 33% , 4% , 3% , and 1% of the normal value at 1,2,3,4 weeks respectively. In the α-erys- tallin group, the number of RGCs decreased to 51% , 11% ,7% ,3% of the normal value at 1,2,3,4 weeks respectively. RGCs in both groups were more than those in control group. Conclusion Intraocular injection with α-erystallin after optic nerve injury can protect RGCs from injury and cell death.
