摘 要:目的:观察石吊兰醇提取液对荷S180实体瘤小鼠体内抗肿瘤作用,分析对其免疫功能的影响。方法:实验于2003-02/08在赣南医学院动物实验中心完成。取昆明种小鼠90只。先设正常对照组15只,其余小鼠右腋皮下接种瘤液0.2mL,24h后随机数字表法分为模型组、石吊兰2,4g/kg组、环磷酰胺组、环磷酰胺+石吊兰组。正常对照组、模型组用生理盐水腹腔注射,每次20mL/kg,石吊兰2,4g/kg组、环磷酰胺组、环磷酰胺+石吊兰组分别给予石吊兰2,4g/(kg·d),环磷酰胺0.3g/(kg·d),石吊兰2g/(kg·d)+环磷酰胺0.3g/(kg·d),腹腔注射,1次/d,连续10d。第11天称质量后拔眼球取血,采用双抗体夹心ELISA法测定血清白细胞介素2浓度;剥取肿瘤、胸腺、脾组织,计算抑瘤率、胸腺指数、脾指数。结果:①石吊兰4g/kg组、环磷酰胺组和环磷酰胺+石吊兰组对肿瘤组织有明显的抑制作用(42.1%,74.7%,71.3%,P<0.05~0.01);环磷酰胺组和环磷酰胺+石吊兰组抑瘤作用优于石吊兰4g/kg组(P<0.01)。②荷瘤小鼠胸腺指数、脾指数均低于正常小鼠(P<0.05~0.01),但石吊兰4g/kg组胸腺指数、脾指数均较模型组显著提高,差异有显著性意义(P<0.01),石吊兰2g/kg组仅胸腺指数高于模型组(3.84±0.21,P<0.01)。环磷酰胺+石吊兰组能提高小鼠胸腺指数与脾指数,与环磷酰胺组比较,差异有显著性意义(2.51±0.33,1.69±0.15;5.03±0.51,4.30±0.27;P<0.01)。③石吊兰2,4g/kg组小鼠血清中白细胞介素2含量较正常对照组和模型组均显著提高,差异有显著性意义[(253.3±17.8),(330.9±24.2),(189.1±45.3),(145.6±15.7)ng/L,P<0.01]。环磷酰胺+石吊兰组白细胞介素2含量高于环磷酰胺组[(277.1±33.2),(133.3±12.5)ng/L,P<0.01]。结论:石吊兰醇提取液具有抑制S180实体瘤生长及提高荷瘤小鼠免疫功能的作用,可能与其提高血清中白细胞介素2水平有关。AIM: To observe effect of Lysionotus Pauciflorus Maxim alcohol extract (LAE) on anti-sarcoma S180 in mice and analyze the effect on immunological function. METHODS: The experiment was completed in the center of animal experiment in Gannan Medical University from February to August 2003. Totally 90 Kunming mice were selected in this study. Fifteen mice were regarded as normal control group. Other 75 mice were inoculated subcutaneously with 0.2 mL liquid containing tumor cells in right axilla. After 24 hours, mice were randomly divided into 5 groups: model group, 2, 4 g/kg LAE groups, cyclophosphamide group, cyclophosphamide plus LAE group. The mice in the model group and normal control group received saline by intraperitoneal injection, once 20 mL/kg. The mice in the 2, 4 g/kg LAE groups, cyclophosphamide group and cyclophosphamide plus LAE group received 2, 4 g/kg LAE per day, 0.3 g/kg cyclophosphamide per day and 2 g/kg LAE per day plus 0.3 g/kg cyclophosphamide per day by intraperitoneal injection, once a day for 10 days. At day 11, blood was collected by drawing eyeball after being weighed. The concentration of interleukin (IL)-2 in serum was determined by the method of double antibody sandwich ELISA. Tumor, thoracic gland and spleen tissues were cut to calculate anti-sarcoma rate, thoracic gland index and spleen index. RESULTS: (1)There were significant anti-sarcoma effect in the 4 g/kg LAE group, cyclophosphamide group and cyclophosphamide plus LAE group (42.1% ,74.7% ,71.3% ,P 〈 0.05-0.01). The inhibitory effect was better in the cyclophosphamide group and cyclophosphamide plus LAE group than the 4 g/kg LAE group (P 〈 0.01). (2)Thoracic gland index and spleen index in mice bearing tumor were lower than the normal mice (P 〈 0.05-0.01 ). But the thoracic gland index and spleen index in the 4 g/kg LAE group were increased obviously as compared with the model group, and there were significant differences (P 〈 0.01 ).
