摘  要:目的比较轮状病毒Wa株和SA11株对MA104细胞的感染及复制能力,研究不同毒株对宿主细胞的感染效力。方法差速离心浓缩病毒颗粒,FFA法检测病毒滴度,流式细胞术检测病毒对MA104细胞的感染效率,ELISA检测不同病毒颗粒浓度,并比较2毒株间异同。结果Wa株感染效率随病毒量的变化明显快于SA11株,极限感染效率也较SA11高;2毒株病毒产量均随感染病毒用量增加而明显升高,而感染了SA11细胞的平均病毒生成能力显著高于感染Wa株的细胞。结论轮状病毒Wa株与SA11株感染细胞效力差异显著(P<0.05),为进一步研究轮状病毒感染宿主细胞及与之相互作用特点提供了实验依据。Objective To compare the infectivity and reproductive capability in MA104 cells of different rotavirus strains Wa and SA11, and analyze their differences in infection, Methods The rotavirus was prepared by differential centrifugation, and the virus liters were detected by FFA. Flow cytometry was used to detect virus infectivity to MA104 cells, ELISA was employed to detect rotavirus particle amounts and analyze the differences between the two rotavirus strains. Results The infective ability of Wa to MA104 cells was obviously higher than thatofrotavirusSA11, while the cells infected by SA11 have higher virus productivity than those infected by Wa strain. Conclusion Rotaviruses Wa and SA11 has obviously different infection pattern. The results give a basis for studying the properties of rotavirus interaction with host cells.
