摘  要:选用健康种猪18头,按1、2、3月龄分为3组,每组6头,无菌采取前腔静脉血10 mL,肝素抗凝,分离外周血单个核细胞(PBMC),计数并稀释成1×10^7个/mL、5×10^6个/mL、2.5×10^6个/mL、1.25×10^6个/mL、6.25×10^5个/mL五个浓度梯度;植物血凝素-p(PHA-P)配成100μg/mL、50μg/mL、25μg/mL、12.5μg/mL、6.25μg/mL五个浓度梯度.采用正交试验法在96孔细胞培养板中加入不同浓度的PBMC和PHA-p,用MTT法检测淋转试验结果.三组试验结果均表明PBMC和PHA-p对猪淋巴细胞增殖反应均有显著的影响(0.01<p<0.05),其最佳反应条件组合为:PBMC浓度为1.25×10^6个/mL,PHA-p浓度为12.5μg/mL,同时也表明年龄大小不影响猪的淋转试验.The paper choices 18 caput of health boardistinguishedinto 3 groups by different month (one month, two month, three month).Every group includ 6 caput of health boar; asepsis adopting blood of vein 10ml in former celiac, adding anticoagulant, separating peripheral blood monocyte (PBMC), taking count of PBMC and diluting different 5 grads of chroma (1×10^7 individual/ml, 5×10^6 individual/ml, 2.5×10^6 individual/ml, 1.25×10^6 individual/ml, 6.25×10^5 individual/ml); plant prothrombin-p (PHA-p) was matched different 5 grads of chroma (100μ g/ml, 50μ g/ml, 25μ g/ml, 12.5μ g/ml, 6.25μ g/ml). Adopting orthogonal test PBMC and PHA-p of different chroma entered into 96 apertures cell-cultivated board, deteeting result of lymph transform test by means of MTT. 3 groups of test result indicate: PBMC and PHA-p significantly influenced increment reactivity of lymphocyte of pig (0.01〈p〈0.05), that optimal condition of reactivity: chroma of PBMC is 1.25×10^6 individual/ml, ehroma of PHA-p is 12.5μ g/ml, simultaneity, age magnitude didn't influence lymph transform test of pig.
