摘 要:目的:通过DNA疫苗与痘病毒联合免疫效果的分析,探讨这种免疫方案的优势及跨膜修饰的DNA质粒诱导的特异性CTL强度。方法:用跨膜修饰的DNA质粒PDRVISV1.0-gagtm在0、2、4周初始免疫BALB/c小鼠,在第6周用痘病毒rVV-syngag加强免疫。在第10周采集血清样本进行抗HIV抗体亚型检测,然后处死小鼠检测小鼠的细胞免疫水平。结果:这种联合免疫的策略免疫组诱导出了特异性CTL反应并且抗体亚型检测免疫反应趋向于Th1型细胞免疫。结论:本实验为提高疫苗免疫原性提供了新的免疫方法。为免疫策略的深入研究奠定了一定的基础,提供了一定的数据支持和启示。Objective.To use immunogenic analysis of DNA vaccine combined with recombinant vaccinia virus to study the advantage of the immunity strategy and its specific CTL function induced by transmembrane modified DNA plasmid. Methods: BALB/C mice were immunized with DNA vaccine at weeks 0,2 and 4 and with recombinant poxvirus at 6 week. Blood sample was collected,antibody subtypes against HIV were detected,and mice were killed and cellular immune response was examined by ELISPOT at 10 week. Results:In mice PDRVISV1.0--gagtm/rVV syngag group,the specific CTL functions were detected by EIJISPOT,and the detection of antibody subtypes indicated that the immune response induced by the vaccine had a tendency to Thl--based cellular immune response. Conclusion;Our study provides a new method for improving the immunogenicity of DNA vaccine, and has established a foundation for developing immunity strategy for Chinese HIV,providing data and enlightenment.
