摘 要:分别用改进的CTAB法、SDS法和高盐低pH法从蒜头果种仁中提取基因组DNA,通过琼脂糖凝胶电泳和紫外分光光度法检测所提取基因组DNA的纯度.结果表明:用改进的CTAB法、SDS法和高盐低pH法所提取的蒜头果基因组DNA的OD260/OD280值在1.8~2.0之间,带型清晰无拖尾,说明所提取的DNA保持完好.其中改进的CTAB法所提取的基因组DNA质量最好、纯度也最高,为蒜头果分子生物学研究及下游操作奠定了基础.Three methods of DNA extraction were studied using improved methods of CTAB, SDS and low pH with high salts on the seeds of Malania oleifera. The extracted genomic DNA purity was detected by agarose gel electrophoresis and a U V-vis recording spectrophotometer. The genomic DNA extracted using improved methods of CTAB, SDS and low pH with high salts has good purity with OD260/OD280 ratio between 1.8 and 2.0. The improved CTAB method gave satisfied resuhs,such as the best quality and highest purity of extracted DNA, which was an important basis for further study of molecular biology in Malania oleifera.
