摘 要:目的:探讨C-jun,HSP70在大鼠脊髓损伤组织中的表达,并观察甲基强的松龙(MP)的干预作用.方法:采用Allens打击法建立急性脊髓损伤(ASCI)模型,观察MP组和NS组在伤后1,3,6h,1,2,3d时间点组织中C-jun,HSP70阳性反应物的表达.结果:MP组与NS组脊髓损伤组织中存在C-jun,HSP70的阳性表达.ASCI后C-jun表达增加,3h最为显著,MP组在伤后1,3,6h时间点C-jun的表达明显少于NS组(P<0.05).ASCI后HSP70表达增加,1d最为显著,MP组在伤后6h,1,2d时间点HSP70的表达明显多于NS组(P<0.05).C-jun,HSP70在对照组有少量表达.结论:急性脊髓损伤组织中存在C-jun,HSP70的动态阳性表达,MP可以抑制损伤性因素(C-jun蛋白)的表达,促进保护性因素(HSP70蛋白)的表达,从而减轻继发性脊髓损伤.AIM: To discuss the expressions of C-jun and heat shock protein 70 (HSP70) in injured spinal cord tissues of rats and the interference effects of methylprednisolone (MP). METHODS: Fourty-eight female Wistar rats were randomly assigned to MP and normal saline (NS) groups. The acute spinal cord injury models were made by Allen's method. We observed the expressions of C-jun and HSP70 in each group 1, 3, 6 h, 1, 2 and 3 d after injury. RESULTS: Positive expressions of C-jun and HSP70 were found in the injured spinal cord tissues of NS and MP groups. The expression of C-jun increased after acute spinal cord injury, reached the top at 3 h. The expression of C-jun in MP group 1, 3, and 6 h after injury was lower than that in NS group with statistically significant difference ( P 〈 0.05 ). There was no obvious difference between the 2 groups 1,2 and 3 d after injury. The expression of HSP70 increased after acute spinal cord injury, and get to the maximum at 1 d. The expression of HSP70 in MP group 6 h, 1, and 2 d after injury was higher than that in NS group with statistically significant difference ( P 〈 0. 05 ). There was no obvious difference between the 2 groups 1,3 h and 3 d after injury. There was fewer expression in control groups. CONCLUSION: Positive expressions of C-jun and HSP70 were found in the injured spinal cord tissues. MP can restrain the expression of risk factor ( C-jun protein), and promote the expres- sion of protective factor (HSP70). Therefore, MP can relieve secondary spinal cord injury.
