摘 要:目的研究S-腺苷蛋氨酸(SAM)对人肝细胞和肝星状细胞(HSC)(LX-2细胞株)增殖和氧应激及转化生长因子(TGF)β1表达的影响。方法用不同浓度的SAM培养肝细胞和HSC,用MTT法检测SAM对肝细胞和HSC增殖的影响;用次氮基三乙酸铁(Fe-NTa)和SAM共同培养肝细胞和HSC,检测超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量;用不同浓度的SAM培养HSC,ELISA方法检测HSC表达TGF-β1状况。结果在一定浓度范围内,SAM可抑制HSC增殖,而对肝细胞增殖无影响;SAM可增加肝细胞和HSC的SOD活力,降低其MDA含量,同时可抑制HSC的TGF-β1表达。结论SAM可抑制HSC增殖,保护肝细胞和HSC免受氧应激损伤,抑制HSC分泌TGF-β1,显示其具有抑制HSC增殖、抗氧化和抗肝纤维化作用。Objective To investigate the effect of S-ademetionine(SAM)on the proliferation, oxidative stress and transforming growth factorβ1 (TGFβ1) expression of human hepatocytes and hepatic stellate cells (HSCs)( LX-2cell line). Methods Human hepatocytes and HSCs were incubated with various concentrations of SAM, and the effects of SAM on the proliferation of hepatocytes and HSCs was studied by 3 -(4,5 -dimethyhhiazol-2-yl) -2,5 - diphennylterazolium bromide (MTT) colorimetric assay. Human hepatocytes and HSCs were co-cultured with SAM and ferric nitrilotriacetic acid (Fe-NTa), superoxide dismutase (SOD) activity and malondialdehyde (MDA) contents were detected. HSCs were incubated with various concentrations of SAM, the level of TGF-β1 was detected with ELISA. Results Within a given level range of SAM, the proliferation of HSC was significantly suppressed, but the proliferation of hepatocytes was not affected. SAM obviously increased the activity of SOD and decreased the contents of MDA in hepatocytes and HSCs, and inhibited the expression of TGFβ1 in HSCs. Conclusion SAM can inhibit the proliferation of HSC, protect hepatocytes and HSCs from oxidative stress, and inhibit the secretion of TGFβ1 in HSCs.
