高效液相色谱法测定青黛中靛玉红的含量

摘　　要：目的建立测定青黛中靛玉红的含量方法。方法采用高效液相色谱法，以spherisorb C18（4．6mm×200mm）为色谱柱，甲醇-0．1mol／L乙酸铵（65：35）为流动相，检测波长为280am，流速为1ml／min，柱温35℃。结果青黛中靛玉红的含量在5．98～59．8ug／／ml范围内线性良好（r=0．9995）。平均回收率99．87％，RSD1．31％。结论该方法测定简便快速、可靠，可作为青黛中靛玉红含量的质量控制。Objective To establish the method of determination of indirubin from Indigo Naturalis. Methods HPLC was employed, which chromatographic column was spherisorb C18 （4.6 mm × 200 mm）, mobile phase was methanol-0.1 mol/L of ammonium acetate, flow rate was 1 ml/min, detection wavelength was 280 nm and column temperature was 35℃. Results A good linear eorrelationship was obtained when indirubin content ranged from 5.98 ug/ml to 59.8 ug/ml（r =0. 999 5）. Average recovery was 99.87%, RSD was 1.31%. Conclusion This method is simple, rapid, reliable and suitable for assay of indirubin from Indigo Naturalis.
