摘  要:目的优化HB-Ⅰa脂质体的工艺处方,提高包封率。方法采用逆相蒸发法制备HB-Ⅰa脂质体,以包封率为指标,通过正交实验对工艺处方进行了优化研究,用HPLC法测定其包封率。用透射电镜扫描观察其外观形态和测定平均粒径。结果最佳的处方比例为卵磷脂:胆固醇:HB-Ⅰa=400∶200∶60,平均包封率为(85.92±0.61)%,平均粒径为11.25μm。结论优化的处方工艺合理,包封率显著提高。Objective To optimize the preparation and formulation of HB - Ⅰ a liposome and improve the entrapment efficiency. Methods HB - Ⅰ a Liposomes were prepared by reversed - phase evaporation . Orthogonal design was adopted to screen the formulation of HB - Ⅰ a liposomes on the basis of the entrapment efficiency. The entapment efficiency of HB - Ⅰ a was determined by HPLC . The morphology and particle size of the preparation were examined by transmission electron microscope. Results The op- timum formula was PC:CH:HB - Ⅰ a =400:200:60. The average encapsulation efficiency was (85.92 ±0.61 )%. The average diameter was 11.25um, Conclusion The formulation of HB - Ⅰ a is reasonable and can improve the entrapment efficiency.
