摘 要:目的建立反相高效液相色谱(RP—HPLC)法测定银黄颗粒制剂中绿原酸含量的方法及测定不同厂家银黄颗粒制剂中绿原酸的含量。方法采用RP—HPLC法检测银黄颗粒制剂中绿原酸的含量,色谱条件:Kromasil C18色谱柱,以乙腈-甲醇-0.4%磷酸水溶液(含0.6%三乙胺,5:15:80)为流动相,流速1ml/min,检测波长327nm,柱温为室温。结果绿原酸在6.23×10^-3~100.50×10^-3mg·ml^-1范围内与峰面积呈良好的线性关系(r=0.9998),平均回收率为98.30%。RSD=2.35%(n=6)。结论所建立的色谱方法可以简便、准确测定银黄颗粒中绿原酸的含量,重现性较好。各厂家生产的银黄颗粒中绿原酸含量差别较大,有必要建立绿原酸的定量控制方法。Objective To establish an HPLC method and determine the content of chlorogenic acid in Yinhuang Granules from different pharmaceutical factories. Methods RP - HPLC was used to determine the content of chlorogenic acid in Yinhuang granules . The column was Kromasil C18- The mobile phase was nitrile - methanol - water (containing 0. 6% triethylamine ,5: 15: 80) , the detection wavelength was 327 nm. The column temperature was room temperature. Results There was a good linear relationship between the concentration of chlorogenic acid and absorption area value in range of 6. 23 × 10^-3 - 100. 50 × 10^-3 mg · ml^-1, r = 0. 999 8. The average recovery was 98. 30%, RSD = 2.35% (n = 6). Conclusion The chromatographic method established can be used simply and accurately to determine the content of chlorogenic acid in Yinhuang granules and it has good repeatability.
