Effect of Extraneous Gibberellin(GA_3) on Astaxanthin Accumulation of Haematoccus pluvialis

摘  要:在对数生长期的藻液中分别加入不同质量浓度的赤霉素(GA3)溶液后,立即进入胁迫培养(25℃,5000 lx,24 h连续光照+营养盐饥饿),诱导细胞合成积累虾青素。通过显微观察和定期取样测定虾青素含量。试验结果表明,添加5.0 mg/L GA3处理组的细胞比对照组提前14 d完全变红,虾青素产量比对照组提高64.8%,达15.9 mg/L;20.0 mg/L GA3处理组细胞比对照组提前20 d完全变红,但虾青素产量与对照组相比下降19.0%。Effect of extraneous GA3 on astaxanthin accumulation was studied in Haematoccus pluvialis. Different concentrations of GA3 were added into the vegetative'green cells. H. pluvialis cells were cultivated under stressful conditions (24 h light period, 5000 lx, 25 ℃, nutrition deficiency) to promote astaxanthin accumulation. In this stage, morphological changes in cells treated by different concentration grades of GA3 were observed with light microscopy. Moreover, astaxanthin and chlorophyll contents were "determined with absorption spectral (OD490 and OD673, respectively). The results showed that astaxanthin accumulation was promoted at 5.0 mg/L GA3 in large scale. H. pluvialis cells treated by 5.0 mg/L GA3 completed astaxanthin accumulation advanced 14 days ahead in the control and astaxanthin production of H. pluvialis cells was 64.8% higher than that of the control. H. pluvialis cells treated by 20.0 mg/L GA3 completed astaxanthin accumulation 20 days ahead of the control. But astaxanthin production of H. pluvialis cells was 19% lower than that in the control.
