滁菊组织培养脱病毒技术研究

摘　　要：采用改良热处理结合茎尖分生组织培养的方法,建立滁菊组培脱毒快繁技术体系。研究结果表明,滁菊茎段诱导的最佳培养基为MS+2.0 mg/L6-BA+0.1 mg/LNAA;茎尖增殖的最佳培养基为MS+1.0 mg/L6-BA+0.1 mg/LNAA;壮苗生根的最佳培养基为1/2MS+0.1 mg/LNAA。接种脱毒结果为,脱毒率>83%,茎尖成活率>42%。Rapid propagation technical system of the virus--free plantlets of Chuzhou Chrysanthemum was established by shoot tip meristematic tissue culture combined with improved heating treatment, The optimum stem induction medium was MS ＋ 2.0 mg/L 6--BA ＋ 0.1 mg/L NAA. The optimum shoot tip propagation medium was MS ＋ 1.0 mg/L 6--BA ＋ 0,1 mg/L NAA, The optimum rooting medium was 1/2 MS ＋0.1 mg/L NAA, The virus-free ratio of shoot tips was above 83 % and the survival ratio was above 42 %.
