摘  要:目的建立实验性变应性脑脊髓炎(EAE)动物模型,为进一步研究其发病机制提供依据。方法用髓鞘碱性蛋白(MBP)加福氏完全佐剂免疫诱导BALB/c小鼠,观察和比较动物的神经系统症状和病理变化,ELISA方法检测脾细胞培养上清中干扰素-γ(IFN-γ)的水平。结果实验小鼠出现EAE的临床症状,光镜下见血管周围炎性细胞浸润,呈袖套样改变;EAE组的IFN-γ的水平明显高于对照组(P<0.05)。结论本实验所构建的EAE模型稳定、方法可靠。Objective To construct animal model for further elucidating the pathogenesis of experimental allergic encephalomyelitis (EAE). Methods The BALB/c mice were injected with myelin basic protein together with complete Freund's adjuvant, and the neural symptoms and pathological changes of the treated mice were observed. IFN -γ level was detected by ELISA. Results EAE clinic symptoms were observed in most of the MBP treated mice. Sleevelet- like inflammatory cells infiltrated surrounding blood vessel had also been discovered by microscope. The EAE models showed higher levels of IFN -γ than the control groups. Conclusion The construction of EAE animal model was stable and reliable.
