摘  要:目的观察黄芪多糖(Astragalus Polysaccharide)与白芍总苷(total glucosides of paeony)及其配伍对THP-1巨噬细胞源性泡沫细胞内脂质的影响,从抑制巨噬细胞内脂质聚集角度探讨中药有效成分的抗动脉粥样硬化机制。方法应用显微镜摄像技术和图形分析软件半定量测量无apoA-I培养基/apoA-I培养基中THP-1巨噬细胞源性泡沫细胞内脂质的含量。结果无apoA-I培养基中黄芪组、配伍组泡沫细胞内胆固醇脂质染色面积比对照组明显增多(P<0.05);apoA-I培养基中白芍组、配伍组胆固醇脂质染色面积比对照组明显减少(P<0.05)。结论黄芪多糖可增强对氧化型低密度脂蛋白的吞噬能力,白芍总苷能够促进细胞内胆固醇外流。黄芪多糖和白芍总苷配伍(12∶1)有潜在的抗动脉粥样硬化作用。Objective To observe the effects of Astragalus Polysaccharide (ASP) and total glucosides of paeony (TGP) on lipid accumulation in THP- 1 macrophage- derived foam cells. Methods The content of lipid accumulation in THP- 1 macrophage - derived foam cells was measured through photoes under microscope and graph analysis software. Results In the culture medium without apoA - I, the content of lipid accumulation in the groups of ASP was increased significantly ( P 〈 0. 05) than that in the controlled groups, while under the culture medium with apoA - I, the contents of lipid accumulation in the groups of TGP and TGP combined ASP were decreased significantly ( P 〈 0. 05) than that in the controlled groups. Conclusions ASP may increase the phagocytic ability of ox - LDL, TGP may promote the cholesterol outflow. TGP combined with ASP seem to have potential therapeutic effects to atherosclerosis.
