清金得生片对人肺腺癌细胞端粒酶及凋亡相关基因蛋白表达的影响

摘　　要：目的探讨清金得生片抑制肺癌细胞生长的相关分子机制。方法采用免疫组织化学染色(SABC)方法检测清金得生片含药血清处理后人肺腺癌细胞株端粒酶活性和凋亡相关基因bcl-2、bax的表达水平。结果清金得生片含药血清作用72h,人肺腺癌细胞端粒酶活性明显抑制,同时可下调bcl-2蛋白阳性细胞染色率、上调bax蛋白阳性细胞染色率。结论清金得生片通过抑制人肺腺癌细胞端粒酶活性,调节bcl-2和bax蛋白的表达,发挥抗肿瘤作用。Objective To explore the molecular mechanism of Qingjin Desheng Tablets on inhibiting the growth of lung cancer cells and to supply experimental evidences for Chinese herbal medicine preventing and curing lung cancer in clinical application. Methods Immunohistochemistry staining was used to detect the activity of telomerase and the expression of bcl - 2 and bax in human lung adenocarcinoma cells after treated with serum containing Qingjin Desheng Tablets. Resuits The activity of telomerase was inhibited significantly and the positive rate of bcl - 2 protein reduced, and the positive rate of bax2 increased in human lung adenocarcinoma cells after 72 hour- treatment with the serum containing Qingfin Desheng Tablets. Conlusion Qingfin Desheng Tablets exert the anti - tumor action through inhibiting the activity of telomerace and modulating the expression of bcl - 2 and bax protein. adenocarcinoma cells; Telomerase; Bcl- 2; Bax ; Immunohisto- chemistry
