问荆合剂对类风湿性关节炎模型大鼠血清TNF-α的影响

摘　　要：目的探讨问荆合剂对类风湿性关节炎(rheumatoid arthritis,RA)动物模型的疗效及其作用机制。方法采用弗氏完全佐剂建立实验性大鼠RA动物模型,将50只大鼠按体重随机分为5组(空白对照组、模型对照组、问荆合剂组、甲氨碟呤组和问荆合剂加甲氨碟呤组)。观察其治疗前后肿瘤坏死因子-α(TNF-α)的变化。结果各治疗组的血清TNF-α水平有显著的降低,以上各组内治疗前后比较有显著性差异(P<0.01);各治疗组与模型对照组比较均有显著性差异(P<0.01);问荆合剂加甲氨碟呤组与问荆合剂组、甲氨碟呤组比较有显著性差异(P<0.05)。结论问荆合剂能明显下调RA模型大鼠前炎性因子TNF-α水平。问荆合剂对TNF-α的调控作用是其治疗RA的作用机制之一。Objective To investigate the therapeutic effect and mechanism of Wenfing Mixture on animal model of rheumatoid arthritis （RA） from the breakthrough point of auto - immunity and inflammatory responses. Methods The RA rat model was established by Freund＇s complete adjuvant（FCA）. The serum TNF -α levels were detected with enzyme -linked immunosorbent assay（ELISA） before and after treatment. Results The TNF- α serum levels in three therapeutic groups （ Wenfing Mixture group, Methotrexate group and Wenfing Mixture ＋ Methotrexate group） were decreased significantly as compared with the model group （ P 〈 0. 01）, and showed obvious differences with pre - treament respectively （ P 〈 0.01）. The differences between Wenjing Mixture ＋ Methotrexate group and Wenfing Mixture group or Methotrexate group were also significant （ P 〈 0. 05）. Conclusions Wenjing Mixture can obviously clown - regulate the serum levels of TNF - α in RA rat model, which may be one of the therapeutic mechanisms of Wenjing Mixture on the treatment of RA.
