TGFβ1介导雌二醇对前列腺间质细胞smoothelin表达的促进作用

摘　　要：研究了雌激素在促进前列腺基质细胞化过程中作用的分子机制,在前列腺基质细胞培养液中分别添加 TGFβ1,雌二醇、雌二醇和 ICI182.780、睾酮、睾酮和芳香化酶抑制剂、双氢睾酮、雌二醇和 TGFβ1中和抗体;用半定量 RT-PCR 的方法检测基质细胞中平滑肌细胞特异蛋白 smoothelin 和 TGFβl mRNA 的表达水平;用ELISA 测定基质细胞培养基中 TGFβ1的浓度.结果显示 TGFβ1可明显促进 smoothelin 的表达;雌二醇和睾酮能明显刺激 TGFβ1基因和蛋白的表达水平,而雌激素受体抑制剂 ICI182.780和芳香化酶抑制剂可分别抑制雌二醇和睾酮对 TGFβ1表达的促进作用;双氢睾酮对 TGFβ1基因的表达没有影响.TGFβ1中和抗体能抑制雌二醇对 smoothelin 的促表达作用.结果显示雌二醇可能通过诱导 TGFβ1促进前列腺平滑肌细胞特异蛋白smoothelin 的表达.To investigate molecular mechanism of estrogen induction of prostatic stromalcells differentiation, the prostatic stromal cells were treated by TGFβ1, estradiol estradiol and ICI182. 780, testosterone, testosterone and aromatase inhibitor, dihydrotestosterone, estradiol and TGFβ1 neutralizing antibody. Then, we investigated smoothelin and TGFβ1 mRNA expression in stromal cells using semi-quantity RT-PCR. And we also studied the TGFβ1 secretion level of the stromal cells by using an ELISA kit. TGFβ1 increased mRNA expression of smoothelin. Estradiol and testosterone can both stimulate TGFβ1 mRNA expression and protein secretion. However, estradiol and testosterone＇s function can be inhibited by ICI182. 780 and aromatase inhibitor, respectively. A neutralizing antibody to TGFβ1 inhibited the stimulation of smoothelin by estrodial. The result showed that estradiol enhanced smoothelin expression by stimulating the TGFβ1 expression and its secretion level.
