摘 要:目的探讨当归挥发油对人脐静脉内皮细胞增殖、凋亡和胶原合成的影响。方法体外培养人脐静脉内皮细胞,用噻唑蓝比色法检测细胞增殖活性,用流式细胞术分析细胞周期及凋亡,用放射免疫法测定胶原合成。结果低浓度(≤4mg/L)当归挥发油促进细胞增殖(P〈0.05),降低G0/G1期细胞且增加S期细胞(P〈0.05),降低凋亡率(P〈0.05),而高浓度(≥16mg/L)时抑制增殖(P〈0.05),增加G0/G1期细胞且减少S期细胞(P〈0.05),增加凋亡率(P〈0.05)。当归挥发油呈剂量和时问依赖性抑制细胞合成胶原(P〈0.05或0.01)。结论当归挥发油对人脐静脉内皮细胞的增殖呈低浓度刺激高浓度抑制的双向调节作用,但对胶原合成呈抑制效应。Objective To investigate the effects of angehcanaphtha on prohferation, cell cycle, apoptosis, and collagen synthesis of human umbilical vein endothelial cells (HUVEC). Methods HUVEC was cultured and passaged in Dulbecco's modified Eagle's medium (DMEM) and treated with angelicanaphtha 1 mg/ L, 4 mg/L, and 16 mg/L at 1, 3, 5, and 7 day respectively. The proliferation was measured with MTT method. The cell cycle and apoptosis were analyzed with flow cytometry and collagen synthesis was determined with radioimmunoassay. Results The proliferation of the HUVEC was accelerated by angelicanaphtha≤4 mg/L and inhibited by angehcanaphtha at 16 mg/L (P 〈 0.05). Lower concentration (≤4 mg/L) of Angelicanaphtha decreased cells in G0/G1 phase and increased significantly cells in S phase and inhibited the apoptosis (P 〈 0.05 ). However, angelicanaphtha at 16 mg/L increased cells in G0/G1 phase and decreased cells in S phase and induced the apoptosis (P 〈 0.05). The collagen synthesis of HUVEC was inhibited by angelicanaphtha in concentration-dependent manner ( P 〈 0.05 or 0.01 ). Conclusion The proliferation effects of angelicanaphtha on HUVEC had dualistic regulation of increase by lower-concentration and inhibition by higher-concentration. Collagen synthesis of HUVEC was inhibited by angelicanaphtha in concentration-dependent manner.
