摘 要:建立了一种测定苦皮藤根皮及其产品中苦皮素A的反相高效液相色谱方法.采用Zorbax eclipse XDB C18(150 mm×4.6 mm i.d.,5μm)色谱柱,以甲醇-水(V甲醇∶V水=70∶30)为流动相,流速为1.0 ml/min,在波长232 nm处进行检测.实验结果表明,在0.000 9~0.020 7 mg/ml范围内,苦皮素A的峰面积与质量浓度呈良好的线性关系,相关系数R=0.999 5,平均回收率为98.8%,相对标准偏差RSD=0.313%.该法简便、快速且重现性好,适用于苦皮藤原料及其产品中苦皮素A的定量分析.A method for the determination of angulatin A in root barks and the products of Celastrus angulatus by reverse-phase high performance liquid chromatography was developed. Zorbax eclipse XDB C18(150 mm×4.6 mm i.d., 5 μm)was chosen as analytical column, methanol-water (70:30, V:V) at a flow-rate of 1.0 ml/min as mobile phase with UV detection at 232 nm. Results of experiments prove that the relationship between peak area and concentration of angulatin A in the range from 0.000 9 to 0.020 7 mg/ml showed a good linearity,the correlation coefficient was 0.999 5, the average recovery was 98.8 %, and relative standard derivation (RSD) was 0.313 %. The method is simple, rapid and reproducible, and suitable for quantitative analysis of angulatin A in the materials and products of Celastrus angulatus.
