摘  要:对真菌农药含孢量的检测过程中容易造成误差的关键环节进行了探讨和验证。试验确定,制备孢子悬浮液时可用吐温.80作润湿剂,以高速组织捣碎机8000r/min搅拌60s可使孢子充分分散,单孢率达98%以上;进行浓度稀释时,应根据预计的含孢量尽量采用较小的稀释梯度间隔和减少稀释次数;点样时应预先固定盖玻片,并用小量程范围的可调微量移液器调节至30~50μl吸取样液点样,得到的计数结果误差较小且稳定性好。试验结果为建立含孢量的标准检测方法提供必要依据。Deviation cide technical concen probably caused by manual operation process in testing spore density of fungal pesti- trates were studied and discussed. Examination showed that Tween 80 was a favorable wetting agent for fungal spores. Spores were dispersed almost absolutely in Tween 80 solution by stirred 60 s at 8000 r/min. The rate of single spore could be up to 98 %. And narrow dilution interval and fewer dilution times, designed according to estimated concentration, of spore suspension were good for the enumeration of spores and recommend in the interest of reducing relative deviation. It was needed to fix cover glass on blood counting chamber before dropping samples of spore suspension. Using microvolume pipette and sucking 30 - 50μl sample could make the sample drop easy to enter into the counting chamber but without surplus liquid around it, and obtain better counting stabilization. The results above were useful for establishment of standard for testing spore density of fungal pesticide technical concentrates.
