血清中连苯三酚气相色谱测定方法的建立

摘　　要：建立了测定山羊血清中连苯三酚浓度的气相色谱法.采用6%的丁二酸乙二醇聚酯（DEGS）为固定液,99%高纯度氮气为载气,柱初温170 ℃,维持3 min后,20 ℃/min升温至190 ℃,维持3 min后降温7 min.进样口温度：280 ℃,FID检测器温度：280 ℃,氮气流速：40 mL/min,氢气：70 mL/min,空气：700 mL/min.结果表明：连苯三酚的血药浓度在16～200 μg/mL内,与其峰面积有良好的线性关系（r=0.9967）,最低检测浓度为10 μg/mL.回收率为80%～98.71%,日内精密度小于3.31%,日间精密度小于5.97%.本法简便,准确,重现性好,回收率高,为检测血液中连苯三酚提供了一种可行、快捷的方法.To establish a method for the determination of the concent ration of pyrogallol in serum by GC. The fixed liquid was 6% polydiethylene glycol succinate（DEGS）, nitrogen as carrier gas, column initial temperature 170℃, after 3 minutes rising to 190℃ at the rate 20℃/min, and holding for 3 minutes, then falling for 7 minutes. The injection chamber was 280℃, and the FID chamber was 280℃. The velocity of gas was： N2 ：40 mL/min, H2 ：70 mL/min, air：700 mL/min. The results showed that the calibration curve was linear in the concentration ranged from 16μg · mL^ -1 to 200μg · mL^ -1 （ r = 0. 9967） . The detection limit was 10μg · mL ^-1 and the method recoveries were from 80% to 98. 71%. The RSD within a day was less than 3.31%and the RSD between days was less than 5.97%. The method is simple and precise, and can be used to detect the concentrations of pyrogallol in goats serum.
