新城疫病毒TL1株NP基因的遗传变异研究

摘　　要：目的分析新城疫病毒TL1株NP基因部分序列,并与代表性毒株进行比较。方法根据GenBank登陆的新城疫病毒NP基因序列,设计了1对引物,用RT-PCR技术对新城疫病毒内蒙古分离株TL1的NP基因进行整体扩增。将扩增产物提纯后克隆入pGEM-T easy载体,通过酶切、PCR和测序进行验证。结果测序拼接得出NP基因的序列长度为1 528 bp,该基因的ORF总长为1 470 bp,编码489个氨基酸。与GenBank下载的14株参考毒株比较NP基因编码区全核苷酸序列和氨基酸序列,发现TL1株与鹅源新城疫NA-1株核苷酸的同源性为99.5%,氨基酸的同源性为99.0%;与鹅源新城疫ZJ1株核苷酸的同源性为97.6%,氨基酸的同源性为99.2%,而与传统疫苗株LaSota核苷酸的同源性为85.7%,氨基酸的同源性91.6%。结论TL1株与鹅源新城疫NA-1株、ZJ1株的同源性极高,它们三者亲缘关系较近,同属于基因Ⅶ型新城疫病毒,该毒株相对于经典的NDV在NP基因上已发生了较大的变异。Objective To analyze the sequence of Nucleocapsid （NP） gene of Newcastle disease virus TL1 strain and compare with that of representative virus strain. Methods A pair of primers were designed and synthesized according to the sequences of Newcastle disease virus from GenBank NP gene of TL1 NDV Inner Mongolia isolated was amplified by RT-PCR using primers and cloned into pGEM-T easy vector, the positive clone was identified by enzyme cutting, PCR and sequencing Results The whole sequence of NP gene was analyzed using DNA Star software . The results indicated that the NP gene with 1 528 bp long had a single opening read frame 1 470 bp in length and coded a polypeptide of 489 amino acids. The sequence analysis demonstrated that 14 reference strains loaded down from GenBank, the result of ORF sequence analysis indicated that TL1 strain exhibited 99.5% homology of the nucleotide and 99.0% homology of the amino acid with reference strain NA-1, 97.6 % homology of the nucleotide and 99.2 % homology of the amino acid with reference strain ZJ1, the homology of the nucleotide compared with LaSota was 85.7% and the amino acid was 91.6%. Conclusion The result indicated TL1, NA-1 and ZJ1 probably had a common origin, they were both belong to the genotype Ⅷ of NDV virus, the mutation of the NP gene had varied largely, compared with classic NDV.
