摘 要:目的对9种拟钉螺进行遗传变异研究,为拟钉螺从基因水平分类提供依据。方法采用微卫星锚定PCR(SSR-PCR)技术对海峡两岸4省7地的拟钉螺基因组DNA进行PCR扩增,根据扩增产物按Nei的方法计算遗传距离,并应用SPSS构建系统进化树。结果各地拟钉螺标本的PCR产物呈多态性,其中台湾邱氏拟钉螺与中国大陆拟钉螺地域株遗传距离为0.7391~0.9130;向氏拟钉螺和秉氏拟钉螺间扩增产物相似性最大,遗传距离为0.2174;神农架拟钉螺与向氏拟钉螺、秉氏拟钉螺的遗传距离为0.3333~0.3913;十堰拟钉螺和洪山拟钉螺之间的遗传距离为0.5556;福建拟钉螺与湖北拟钉螺地域株的遗传距离为0.3913~0.7000;武鸣拟钉螺与湖北拟钉螺地域株的遗传距离为0.6667~0.8889。结论9种拟钉螺在基因水平上至少可以分为4类:1)向氏拟钉螺、秉氏拟钉螺和神农架拟钉螺为一类;2)洪山拟钉螺和十堰拟钉螺为一类;3)武鸣拟钉螺为一类;4)福建拟钉螺为一类。台湾邱氏拟钉螺应为钉螺。Objective To study the genetic variation of 9 populations of Tricula and show the evidence for the classification of Tricula on genetic level. Methods Tricula isolates were obtained from seven areas in China. Their genomic DNA was amplified by the simple sequence repeat-anchored PCR(SSR-PCR) technique, Their genetic distances were measured according to their amplified products and the corresponding dendrogram was constructed. Results The SSR- PCR products were polymorphic. The genetic distance between the Chinese mainland isolates of Tricula and the Taiwan isolate was 0. 7391-0. 9130, The similarity between Tricula hsiangi and Tricula pingi was sufficiently close, and the genetic distance was 0. 2174; The genetic distance between Shennongjia and Wufeng(T. pingi and T. hsiangi) isolates was 0. 3333-0. 3913; The genetic distance between Shiyan and Zhongxiang isolates was 0. 5556; The genetic distance between Fujian and Hubei isolates was 0. 3913-0. 7000; The genetic distance between Guangxi and Hubei isolates was 0. 6667-0. 8889. Conclusion The population of Tricula can be divided into four groups in gene level: 1)isolates from Wufeng (T. pingi and T. hsiangi) and Shengnongjia; 2)isolates from Zhongxiang (T. hongshanensis) and Shiyan; 3)isolates from Wuming; 4)isolates from Fujian (T. fujianensis). And at genetic level, the evidence Tricula chiui was a kind of Ocomelania was proved.
