摘 要:目的:比较了溶胶凝胶法对氧化铝基陶瓷表面改性前、后,其细胞毒性的变化。方法:对热处理前、后的纳米二氧化硅凝胶进行红外光谱分析(IR);制备纳米硅涂层改性陶瓷和未改性陶瓷的细胞培养基浸提液,采用四唑盐比色分析法(methyl thiazoly ltetrazolium assay,MTT法)测定材料对人牙龈成纤维细胞(human gingival fibroblast cells,HGFC)生长的影响。结果:IR分析发现纳米二氧化硅凝胶经热处理后Si-OH弯曲振动吸收峰消失;纳米硅涂层改性陶瓷与未改性陶瓷的细胞相对增值率(relative growthrate,RGR)没有明显的差异(P>0.05),细胞毒性级别(cytotoxical grade,CG)为0~1级。结论:热处理加强了纳米硅涂层微结构中的Si-O-Si网络;用溶胶凝胶法进行纳米硅涂层表面改性的氧化铝基陶瓷没有明显细胞毒性。Objective:To evaluate the cytotoxicity of nano-silica coating with sol-gel process of alumina-based ceramic. Methods: Coating gel after heat treatment was analyzed by infrared spectrum(IR), with gel without sintered as control. Human gingival fibroblast cells (HGFC) were cultivated with extractions of alumina-based ceramic with or without nano-silica coating. Relative growth rate (RGR) was analyzed with a methyl thiazolyl tetrazolium (MTT) assay. Results:It can be seen from the IR picture that bending vibration absorption kurtosis of Si-OH vanished after heat treatment. No significant difference was observed between alumina-based ceramic with and without nano-silica coating. Cytotoxical grade (CG) was 0 to 1. Conclusion: Heat treatment may reinforce Si-O-Si net structure of coating gel. Nano-silica coating with sol-gel process of alumina-based ceramic had no significant cytotoxicity to HGFC.
