摘  要:目的观察木耳多糖对实验性家兔动脉粥样硬化斑块中基质金属蛋白酶-13(MMP-13)表达的影响,从而进一步探讨木耳多糖在动脉粥样硬化中的作用。方法将40只健康雄性新西兰白兔随机分为3组:空白对照组(正常组)8只,每天喂普通饲料;阳性对照组(模型组)16只,每天给普通饲料+1g胆固醇+15%猪油;实验组(预防组)16只,每天给普通饲料+1g胆固醇+1g木耳多糖+15%猪油。16周末所有实验用兔麻醉处死,各组行主动脉粥样硬化病理形态学观察,包括大体形态学观察、光镜观察。根据形态学特点分成稳定斑块和不稳定斑块。通过免疫组织化学、特殊染色及电镜对斑块内的病变进行定量分析。结果特殊染色(Masson特染)可见模型组的粥样硬化斑块中有大量胶原纤维增生,而预防组蓝染的胶原纤维数量较模型组少。S-P免疫组织化学可见模型组中MMP-13在粥样斑块中平滑肌细胞及泡沫细胞中的细胞核及细胞质中呈阳性表达,而正常血管壁表达阴性。预防组与模型组对照结果显示,MMP-13在预防组中的表达较模型组明显减少。结论木耳多糖降低斑块中MMP-13的表达,进一步说明其消退斑块的作用;木耳多糖能够降低胶原纤维的生成,说明其不仅可以降低血脂,还可能抑制胶原纤维的产生从而抑制AS的发生发展。Objective To investigate the effects of auricularia auricula polysaccharide (AAP) on the expression of MMP-13 in rabbit arteriosclerosis (AS) plaques for a better insight to the role of AAP in resolution of AS plaques. Methods Forty healthy New Zealand rabbits were randomized into 3 groups to be on a daily diet of usual food (normal group as blank control,n=8), or usual food + 1 g cholesterol + 15% lard (model group as positive control,n=16), or usual food + 1 g cholesterol + 1 g AAP +15% lard (prevention group as treatment subject,n=16). At the end of 16 weeks, all rabbits were sacrificed by overdose anesthesia and studied for both macropathology and micropathology of aortic AS plaques. The plaques were classified into stable and unstable based on morphology, and further quantified using immunohistochemistry, specific staining and electronic microscopy. Results Specific staining (Massion staining) showed massive proliferation of collagen fibers in AS plaques of the model group, which was much less in the prevention group. As found with SP immunohistochemistry, MMP-13 expression was localized in the smooth muscle cells and foam cells of AS plaques but not in those of normal tissue. Compared with the model group, the prevention group showed significantly lower level of MMP-13 expression. Conclusion AAP appeared to markedly reduce the expression of MMP-13 in AS plaques, whir underlies its role in plaque resolution. Suppression of collagen fiber proliferation by AAP also adds to the potential mechanisms by which AAP prevents the development of AS, in addition to reduction of blood lipids.
