摘  要:目的:系统回顾当前国内外微囊化人工细胞技术的临床应用研究进展。资料来源:检索PubMed数据库中1980-01/2006-06有关人工细胞微囊技术进展的文章,检索词“microcapsule,artificial cell,transplantation”,并限定文献语言种类为English。同时检索中国知网医学文献数据库1994-01/2006-06的相关文章,检索词为“微囊化,人工细胞,移植”。资料选择:纳入标准:①人工细胞微囊的成囊材料、成囊工序及微囊发生器的研究。②人工细胞微囊生物特性的研究。③临床应用的研究。④动物实验。排除标准:①较陈旧的文献。②重复研究。资料提炼:共收集到符合上述要求的文献80篇,58篇符合纳入标准。其中研究内容相似的,以近3年内发表在较权威杂志者优先。排除的20篇为较陈旧的文献及重复研究;对符合标准的38篇文献进行分析。资料综合:适当的囊材、先进的制作工序和机械设备,能显著提高微囊的生物相容性、机械稳定性、免疫隔离效果,提高细胞移植的成功率。微囊化技术可明显提高移植细胞在体内的存活时间。微囊化人工细胞移植在甲状旁腺功能低下症、帕金森病、脊髓损伤、急性肝衰竭、肿瘤等疾病的治疗方面取得了良好的效果。结论:微囊化技术可以有效的避免免疫排斥反应对移植细胞的损伤,日益成熟的人工细胞微囊技术在临床疾病治疗方面的广阔的应用前景。OBJECTIVE: To systematically review the research development in the application of micrecapsulated artificial cell technique at home and abroad in recent years. DATA SOURCES: A computer-based online search of PubMed database was undertaken to identify English articles about the advancement of microcapsulated artificial cell published from January 1980 to June 2006 by using the key words of "micrecapsule, artificial cell, transplantation". Meanwhile, we searched CNKI for Chinese relevant articles published between January 1994 and June 2006 with the same key words in Chinese. STUDY SELECTION: Inclusive critena: (1)about the encystations material, process and mechanical equipment of artificial cell micrecapsule; (2)relevant to the biological characteristics of artificial cell microcapsule; (3)clinical application; (4)animal expenment. Exclusive criteria: (1)obsolescent articles; (2)repetitive articles. DATA EXTRACTION: Totally 80 relevant articles were collected and 68 of them were selected in accordance with the inclusive cnteda. Researches with similar contents published in authoritative journals in the past 3 years were firstly selected. Twenty repetitive and old articles were excluded, and 38 literatures in accordance with the criteria were analyzed. DATA SYNTHESIS: Proper matenal, advanced manufacture process and mechanical equipment can significantly enhance the microcapsules biocompatibility, mechanical stability, immunopretective effect, and raise the successful rate of cellular transplantation. Microcapsule technology can significantly increase cell survival in vivo. Micrecapsulated artificial cells have been successfully applied in treating hypothyroidism, Parkinson disease, spinal cord injury, acute liver failure, tumor and so on. CONCLUSION: Encapsulating cell micrecapsules technique is an effective method for immunopretection. The development of artificial cell microcapsules technique will open a promising approach for treating clinical diseases.
