摘 要:目的探讨新生鼠缺氧缺血损伤脑组织水通道蛋白-4(aquaporin-4,AQP-4)表达的变化及意义。方法健康7d龄SD大鼠共80只,分为对照组(40只)和缺氧缺血性脑损伤模型组(HIBD组)(40只)。HIBD组经右侧颈总动脉结扎后,吸入8%O21h,建立HIBD模型。对照组仅行假手术,不予颈总动脉结扎和缺氧。两组均于HIBD模型制成后0h,6h,24h,48h和72h分别处死动物(各时间点动物8只),进行脑含水量观察,Western蛋白免疫印迹检测脑组织AQP-4表达。结果HIBD组6h,24h,48h和72h脑组织含水量均较对照组增加,差异有显著意义(P<0.05);与对照组比较,HIBD组脑组织AQP-4表达在48h内呈下降趋势,72h出现恢复,但仍低于对照组(P<0.05)。结论AQP-4表达下降可能与脑水肿的发生有关,AQP-4可能参与脑内渗透平衡重建。Objective To explore the expression of AQP-4 in the brain tissue of newborn rat with HIBD. Methods eighty 3 ~ 4 day old SD rats were subjected to the ligation of right carotid artery (ischemia), then they were put into a box full with 8% oxygen and 92% nitrogen for an hour (hypoxia). The 3~4 day old SD rats with HIBD model were divided into 5 groups, including 0, 6 h,24 h,48 h and 72 h after setup of HIBD. The normal control groups were also divided into 5 groups same as HIBD groups, the Western blot to detect the AQP-4 expression in the brain tissue of both HIBD and the control groups. Results The brain water content was increased markedly in HIBD groups after 6 h of HIBD and increased further with time in neonatal rats compared with that in the control group. The expressions of AQP 4 were significantly decreased in HIBD groups during the period of 6 h~72 h of HIBD compared with the control group in the SD neonatal brain tissue (P〈0. 05). Conclusion It is likely that the decreased expression of AQP-4 is associated with brain edema in HIBD and in such brain edema conditions AQP-4 may play an important role in the re-establishment of brain osmotic equilibrium.
